Project description – Summary – 3rd funding period

HIV-1 capsids enter the central channel of the nuclear pore complexes (NPCs) and rupture following nuclear import, when the viral genome integrates into the cellular chromatin. Our mechanistic understanding of these steps remain incomplete, although they are pivotal for understanding how the virus becomes an integral part of the host genome cannot be targeted by current therapies. Here we will investigate the spatiotemporal regulation of capsid rapture and genome integration, taking into account physical forces and the crowded environment encountered by the virus during nuclear import and at the level of chromatin.

HIV-1-Kapside werden durch Kernporenkomplexe (NPC) in den Zellkern importiert und brechen danach auf um das virale Genom zu entlassen, welches in das zelluläre Chromatin integriert wird. Unser mechanistisches Verständnis dieser Schritte ist nach wie vor unvollständig, obwohl sie von zentraler Bedeutung sind, um zu verstehen wie das Virus zu einem integralen Bestandteil des Wirtsgenoms wird, welches im Moment nicht therapierbar ist. Wir wollen die räumlich-zeitliche Regulierung des Zellkerneintritts des Kapsid und der Genomintegration untersuchen. Dabei werden die physikalischen Kräfte und die beengte Umgebung berücksichtigt auf die das Virus beim Kernimport und auf der Ebene des Chromatins trifft.
